摘要
目的 :比较三种方法扩增的外周血细胞因子诱导的杀伤细胞 (CIK细胞 )在增殖能力、细胞表型和细胞毒作用三方面的差异。方法 :用三种不同因子组合扩增CIK细胞 ,即IL 2组 :CD3单抗、IFN γ和IL 2 ;IL 1加IL 2组 :CD3单抗、IFN γ、IL 1和IL 2 ;IL 12组 :CD3单抗、IFN γ和IL 12。MTT法测定细胞增殖力 ,流式细胞术分析细胞表型 ,LDH释放法测定细胞毒作用。结果 :三种方法均可使细胞增殖能力显著增加 ,其中IL 12组促增殖能力及CD3+ 细胞所占比例低于另外两组 ,而CD5 6 + 细胞则多于另两组。IL 1和IL 2联合作用组CD4 + 细胞百分率高于其他两组 (P <0 .0 5 )。三种方法扩增的CIK细胞细胞毒性作用无明显差异。结论 :三种方法扩增的CIK细胞均可用于过继免疫治疗的研究。
Objective: To study the cellular prolifer ation,surface antigen expressi on and cytotoxic activity of CIK cells generated by three methods. Meth ods: In addition three methods of CIK cells gereration,IFN-γ and CD3 McAb,IL-1 and IL-2、IL-2、IL-12 were added respectively. Surface antigen expression was detected by flow cytometry,cellular proliferation and cytotoxic activity were analyzed using MTT and LDH release assay.Results: Cells generated by three methods showed high prolife ration activity,though IL- 12 led to lower proliferation activity and number of CD3-positive cells than ot h ers,it could lead to higher percentage of CD56-positive cells.IL-1 addition co ul d increase the number of CD4-positive cells.No significance was found among thr e e groups in cytotoxic activity.Conclusion: All methods mentio nee above can be used to generate immuno-effector cells for immunotherapy.
