摘要
目的 探讨中药泽泻对尿草酸钙结石形成的影响。 方法 采用种晶技术检测泽泻水溶性提取物 (C1、C2 组 )、5 0 %甲醇提取物 (D1、D2 组 )和 10 0 %甲醇提取物 (E1、E2 组 )对体外一水草酸钙晶体生长的抑制指数 (I .I)。 80只Wistar大鼠随机分为 8组 ,对照组 (A组 )、成石组 (B组 )、泽泻水溶性提取物组 (CL、CH 组 )、5 0 %甲醇提取物组 (DL、DH 组 )、10 0 %甲醇提取物组 (EL、EH 组 ) ,分别予不同浓度的泽泻水溶性提取物和甲醇提取物。饲养 4周后 ,检测各组大鼠血生化 ,2 4h尿草酸(Ox)、Ca2 + 、Mg2 + 分泌量和肾组织Ca2 + 、Mg2 + 含量。镜下观察肾组织切片中草酸钙结晶及肾小管扩张情况。 结果 C1、C2 、D1、D2 、E1、E2 组I.I分别为 90 .5 6 %、76 .4 7%、74 .38%、86 .4 1%、6 1.17%和 11.89%。动物实验A组血BUN、Cr明显低于其他各组 ,差异有显著性意义 (P <0 .0 5 ) ;各组间血Ca2 + 、P浓度无显著性差异 ;2 4h尿Ox、Ca2 + 和Mg2 + 分泌量各泽泻实验组与B组差异无显著性意义(P >0 .0 5 ) ;CH、DL、DH 组肾组织Ca2 + 含量明显低于B组 ,差异有显著性意义 (P <0 .0 5 ) ,各组间肾组织Mg2 + 含量差异无显著性 (P均 >0 .0 5 )。A组肾小管正常 ,B组肾小管腔可见大量成片草酸钙结晶存在 。
Objective To investigate the effect of Alisma Orientatis,a Chinese herbal medicine,on urinary oxalate calcium (CaOx) stone formation. Methods The study was carried out both in vitro and in vivo.In the former,aqueous extract from Alisma Orientatis (TAq-ext)(group C 1 and C 2),50% methanolic extract (TMe-ext)(group D 1 and D 2) and 100% TMe-ext (group E 1 and E 2) were tested for the inhibiting index (I.I) on CaOx crystal growth by seeded crystalization technique.In the in vivo study,80 adult male Wistar rats were assgined into 8 groups (n=10 each):group A as normal control;group B,the stone formation;group C L and C H,the TAq-ext;group D L and D H,the 50% TMe-ext and group E L and E H,the 100% TMe-ext.After 4 weeks,the serum BUN,Cr,Ca 2,P,the 24 h urinary excretion Ca 2,Oxalate(Ox),Mg 2 and the renal tissue Ca 2,Mg 2 were detected.All kidneys were removed and examined microscopically for possible CaOx crystals and tubular dilatation. Results The I.I in C 1,C 2,D 1,D 2 and E 1,E 2 groups were 90.56%、76.47%、74.38%、86.41%、61.17% and 11.89%,respectively.In the experimental animal group A,blood BUN and Cr were significantly lower than the others (P<0.05).Blood Ca 2 P,and Ox,Ca 2 and Mg 2 in 24 h urine were similar in all groups of Alisma Orientatis and group B(P>0.05).The renal tissure Ca 2 in C H,D L and D H was significantly lower than that in group B(P<0.05).No significant difference of Mg 2 in renal tissue betweer different groups was noted (P>0.05).Renal tubules were normal in group A;large amount of calcium oxalate crystals could be seen in the lumen of the tubules which were evidently delated in group B.In group C L and C H,scattered calcium oxalate crystals could be seen and the tubules were slightly dilated;in D L and D H,only slight amount of seatered calcium oxalate crystals could be detected with no definite dilatatin of the tubules;in E L and E H groups,the finding was similar to that in group B. Conclusions 50% TMe-ext significantly decreases the Ca 2 in renal tissure and diminishs the tubule dilatation in the in vivo experiment and inhibits urinary CaOx stone formation in the in vitro study.So,the dominant part of Alisma Orientatis on inhibiting CaOx stone formation is the 50% TMe-ext.
