摘要
目的 探讨端粒酶活性与上皮性卵巢癌细胞系化学敏感性的关系。方法 使用顺铂作用于敏感细胞株A2 780及耐药细胞株AD60。在四甲基偶氮唑盐 (MTT)法测定耐药倍数的基础上 ,应用聚合酶链反应 聚丙烯酰胺凝胶电泳 (PCR -PAGE)方法测定端粒酶活性。结果 经MTT测定卵巢癌耐药细胞株AD60的耐药倍数为 2 4。A2 780端粒酶活性随药物浓度的递增明显降低 ,而AD60降低不明显。结论 端粒酶活性与上皮性卵巢癌细胞对细胞毒性药物的化学敏感性相关。
Objective To evaluate the relationship between telomerase activation and chemosensitivity in epithelial ovarian cancer cell lines.Methods The human epithelial ovarian cancer drug-sensitive cell line A2780 and drug-resistance cell line AD60 was treated with cisplatin.MTT was used to assary the multiple of drug resistance.Telomerase activation was detected by polymerase chain reaction-acrylamidegel electrophoresis (PCR-PAGE).Results The multiple of drug resistance of the ovarian cancer cell lines AD60 was 2.4.with the increasing cisplatin concentrations of the telomerase activation of A2780 decreased more significantly than those of AD60.Conclusion The current study suggests that telomerase activation related to the chemosensitivity in epithelial ovarian cancer.
