摘要
从上海地区栽培的香石竹上分离到一种直径为34~35nm 等轴对称的病毒颗粒,在SDS-聚丙烯酰胺凝胶电泳中病毒外壳蛋白分子量为40000道尔顿,并由约372个氨基酸残基组成。病毒核酸经 DNase 1、RNase 1和核酸酶 S1等酶切试验证明为单链 RNA,在1.0%的琼脂糖凝胶电泳中病毒核酸呈现一个分子量为1.2×10~6道尔顿的基因组分和一个较弱的分子量为0.5×10~6道尔顿的组分。根据病毒上述的理化特性以及免疫双扩散实验,证明目前在上海地区分离获得并引起本地区香石竹主要病毒病害的是香石竹斑驳病毒。
An icosahedron virus with a diameter about 33~35nm was isolated from car- nation diseased plants in Shanghai area.The SDS-PAGE demonstrated that the virus con- tains a single capside polypeptide band with molecular weight about 40kd.The amino acids composition analysis indicated that the protein contains 372 amino acids residues.The nucle- ic acid extraction from purified viral preparations showed one band with M.W.approximate- ly 1.2×10~6 and another minor band with M.W.about 0.5×10~6 daltons.This result was confirmed by direct electron microscopic observations revealing two kinds of RNA molecules with different lengths of 2.5×10~4 (?) and 1.6×10~4 (?) respectively.The experiments of RNA 1,DNase 1 and Sl digestions and of the thermal denaturation suggested that the viral genome is a single-stranded RNA.The immunological double diffusion test confirmed that the virus,which causes the main disease of carnation in Shanghai area is the carnation mottle virus.
