摘要
应用异型双功能基化合物N-羟琥珀酰亚胺碘乙酸酯(NHSIA)与蓖麻毒素(Ricin)反应,产生碘乙酰毒素,再与暴露巯基的单克隆抗体(McAb)-PDP反应,通过Sephadex G-200及Sepharose-4B分离,得到缺乏半乳糖结合点免疫毒素。该结合物对人肝癌细胞株Bel7404具有良好的导向杀伤作用,在10^(-9)mol/L浓度时杀伤率为44.1%,非常显著地高于对非靶细胞HeLa的杀伤力(P<0.01)。在10^(-7)mol/L浓度时,对HeLa的杀伤率为39.7%,0.1mol/L半乳糖能够降低这一杀伤作用。
In the present study, heterobifunction linker N-hydroxysuccinimidyl ester (NHSIA) was used to react with ricin to form iodoacetylated ricin and further treated with thiolated monoclonal antibody Hep 1 (McAb-PDP). The reactive product was pu rif icated by passing through Sephadex G-200 and Sepharose 4B columns and an immu notoxin lacking galactose-binding sites was thus obtained. The cytotoxic effect of this conjugate on human liver carcinoma cell line Bel 7404 was investigated in vitro. The experimental result indicated that it had a good cytotoxic activity to Bel 7404 with a killing rate of 44.1% at a concentration of 10-9Mol/L, much greater than that to non target cells(human uterocervical carcinoma cell line Hela)(p<0.01). It also had a killing rate of 39.7% to Hela cell at a concentration of 10-7Mol/L, which could be abolished when 0.1Mol/L galatose was added.
