肿瘤细胞诱生的巨噬细胞细胞毒因子理化性质的分析

STUDIES ON PHYSICOCHEMICAL CHARACTERISTICSOF MACROPHAGE-DERIVED CYTOTOXIC FACTORINDUCED BY TUMOR CELLS

摘要 从K_(562)等肿瘤细胞激活的小鼠腹腔巨噬细胞的培养物中可收集到具有杀伤多种肿瘤细胞的细胞毒样的可溶性因子(简称Mφ-CF)。该因子经56℃、20分钟处理后仍保持活性;在pH7~10时性质稳定。经还原剂(2-巯基乙醇、二硫苏糖醇)、氧化剂(过碘酸钠)及胰酶处理后其活性降低,但不受蛋白酶抑制剂——苯甲基磺酰氟、8-羟基喹啉的影响,SDS-PAGE测得其分子量约为54Kd。本文实验观察到肿瘤细胞诱生的这种Mφ-CF的某些特性与肿瘤坏死因子(TNF)很类似,可能是一种具有二硫键的含糖蛋白分子。 A cytotoxic factor derived from activated macrophages (Mφ-CF) was obtainted by using different tumor cell lines. The Mφ-CF showed a significant cytotoxic effect to a series of tumor target cells. This factor retained its activity after treatment at 56℃ for 20 min. It was stable in pH 7-10. The cytotoxic activity of Mφ-CF was significantly reduced after treatment with trypsin, dithiothreitol, 2-mercaptoethanol and sodium periodate but was not inhibited by protease inhibitors(phenvmethysul fluoride, 8-hydroxyquinine). The molecular weight of this factor was estimated to be 54 Kd by SDS-PAGE. Taken together, these characteristics of the Mφ-CF were quite similar to those of tumor necrosis factor (TNF).
