摘要
反抑制T细胞(TCS)借助辅助T细胞对抗T抑制细胞的抑制效应参与免疫调节。小鼠和人体TCS都表达长绒毛野豌豆凝集素受体(VVR)。而人体TCS则主要是CD8VV^+T亚群,和临床多种疾病免疫功能异常密切相关。 本文用抗VV抗体单色和双色免疫荧光法和流式细胞术(FCM)测知正常人VV^+T亚群的比例是16.6%±2.9%,CD8VV^+T亚群比例是8.45%±1.92%,各种VV^+细胞亚群的功能测定,显示健康人CD8VV^+T细胞亚群,具有拮抗ConA诱导的抑制细胞对杀伤细胞的抑制作用,以及具有增强PBL对PPD诱导的增殖反应,具有正调节功能,而CD8VV^-却具有相反的抑制作用。在后一系统中CD8VV^+细胞相对反应值可高达223.2%,CD8VV^-的相对反应值仅为34.5%。如果在PBL增殖反应中加入CD8VV^-亚群诱发免疫抑制后再加入CD8VV^+亚群可以看到低反应迅速回升,回升幅度和递增加入的CD8^+细胞数量相对应,表明人体CD8VV^+T亚群具有反抑制功能。
Contrasuppressor T cells (TOS) are resistant to the effects of T suppressor cells. It has been found that both human and mouse TOS express receptor of Vicia villosa lectin (WE). Human effective TCS are mainly CD 8 VV+T cell subset which are closely associated with abnormalities of immune activity of many diseases.Routine and double immunofluorescence flow cytometry techniques applied in our experiments showed that the percentage of VV+ T cell subset in normal persons was 16.6±2.9%, and CDVV+ was 8.45±1.92%.Function studies of different VV+ T cell subset revealed that CD8 VV+ T cell in healthy subjects had a contrassupprsssive effect against suppressive activity on the cytotxic ability of NE cells by OonA-indueed suppressor cells. It could also enhance the PPD-induced proliferation of PBL. This fact showed the positive regulatory roles of CD8VV+ cell. In PPD-induced lymphoproliferation in which irradiated CD8VV+and CD8VV+subsets were added as regulators, the relative responses were 223.2% and 34.5%, respectively. In a more sophisticated testing system in which the CD8VV+ and CD8VV+ were combined to creat a more remarkable condition of immune low-responsiveness, adding of the VV+T cells from healthy controls showed typically contrasuppressive activities in a dose-dependent manner.
