血管内皮生长因子及微血管密度与大肠癌浸润、转移及预后的关系 被引量:8

The relationship of vascular endothelial growth factor and micro vascular density with the invasion, metastasis and prognosis of colorectal carcinoma

摘要 目的 探讨血管内皮细胞生长因子 (VEGF)和血管生成与大肠癌发展的关系。方法 采用SABC免疫组化方法 ,检测 6 3例大肠癌、10例癌旁组织及 10例正常大肠组织中VEGF的表达及微血管密度 (MVD)。结果 大肠癌组织中VEGF的表达阳性率明显高于癌旁组织 (P <0 .0 5 ) ,与组织学分级无关 (P >0 .0 5 )。VEGF阳性者MVD值显著高于VEGF阴性者 (P <0 .0 5 ) ,VEGF表达和MVD与大肠癌浸润深度、Dukes分期、淋巴转移和血行转移密切相关 (P<0 .0 5 )。VEGF表达阳性或高MVD的大肠癌患者预后差 (P <0 .0 5 )。结论 VEGF表达与大肠癌血管生成密切相关 ,对大肠癌的生长和浸润转移有促进作用 ,MVD与VEGF可作为大肠癌病程检测的指标 。 Objective To investigate the relationship of vascular endothelial growth factor (VEGF) and angiogenesis with the progression of colorectal carcinoma. Methods VEGF protein expression and microvascular density in 63 cancer tissues, 10 neighboring non-cancerous and 10 normal tissues were examined by SABC immunohistochemical technique. Results The positive rate of VEGF expression in colorectal carcinoma tissues was higher than that of neighboring non-cancerous tissues(P<0.05), MVD was significantly higher in VEGF-positive colorectal carcinoma than in VEGF-negative colorectal carcinoma (P<0.05), but VEGF and MVD were not related to histological grade(P>0.05). MVD and VEGF were positively correlated with the depth of invasion, Dukes stage, lymph node metastasis and blood metastasis of tumor (P<0.05). The prognosis of patients with positive VEGF and high MVD was significantly poorer than that of those with negative VEGF and low MVD (P< 0.05). Conclusion VEGF is closely related to angiogenesis of colorectal carcinoma and promotes growth, invasion and metastasis of colorectal carcinoma. MVD and VEGF can be used as useful markers which guide the choice and better use of therapy, for the stage and prognosis in colorectal cancer.
