摘要
用自己编写的siRNA查找设计的程序sRNAFinder,对来自GenBank的 7个冠状病毒全基因组序列进行了siRNA的查找 ,并分析了各自的分布情况 .结果表明 ,ORF1ab、S、E、M和N这 5个主要基因中 ,候选siRNA占全基因组中所有候选siRNA的比例达到 90 %以上 .根据基因组结构特征 ,对SARS -CoV进行的siRNA分布分析表明 ,在全部 4 4 9个候选siRNA中 ,有 4 4 3个分布于orf1ab中 ,与基于RNAi原理的抗病毒药物设计思想相吻合 .表 8参
RNAi (RNA interference) has been developed as a technique for fundamental research in life sciences and for medicine design in the fields of gene therapy, especially in antiviral infection, such as SARS coronavirus. Scanning, comparing and distribution of candidate siRNAs in the complete genome sequences of 7 coronaviruses from GenBank were accomplished using a compute program, sRNAFinder programmed by authors. The results indicated that more than 90% of the candidate siRNAs were located in the major genes, including orf1ab, S, E, M and N. An analysis according to the structural features of SARS coronavirus genome RNA, 448/454 candidate siRNAs located in orf1ab, suggested that the principle of RNAi-based medicine design might be very different from that of protein target medicine design.
