摘要
目的 研究口服氧化苦参碱对四氯化碳 (CCl4)诱导的大鼠肝纤维化血清生化和纤维化指标的影响。方法 14 0只雄性Wistar大鼠随机分为正常对照组 (n =2 0 )、CCl4模型组 (n =3 0 )、氧化苦参碱预防低剂量组 (n =3 0 )、中剂量组 (n =3 0 )和高剂量组 (n =3 0 )。氧化苦参碱低、中、高剂量组大鼠氧化苦参碱的用量分别为 3 0、60、10 0mg/kg体重 ,每日灌胃 1次 ,共 12周。血清ALT、GGT、总胆红素、白蛋白水平用全自动生化分析仪检测。血清HA、LN、PⅢNP、Ⅳ型胶原含量分别以RIA和ELISA方法检测。肝组织学检查苏木精 伊红染色观察肝组织炎症和纤维化程度。结果 血清学检测结果表明 ,氧化苦参碱预防组较对照组肝功能有明显改善 ,血清肝纤维化指标水平明显降低 ;组织学检查结果表明 ,对照组肝脏炎症和纤维化程度明显高于氧化苦参碱预防组。结论 口服氧化苦参碱可改善CCl4诱导的大鼠肝纤维化血清生化和肝纤维化指标。
Objective To study the influences of oral oxymatrine on serum biochemical and fibrotic indexes of rat liver fibrosis model induced by carbon tetrachloride(CCl 4). Methods 140 male Wistar rats were randomly divided into normal control group(n=20), CCl 4 group(n=30),and oxymatrine-treated group which included low-dose subgroup(n=30),median-dose subgroup(n=30), and high-dose subgroup(n=30). Low, median, and high-dose oxymatrine subgroups were 30,60 and 100 mg/kg·body weight respectively, by daily gastrogavage for 12 weeks. Serum liver function parameters(ALT,GGT,total bilirubin,and albumin) were detected by automatic biochemical analyser, fibrosis indexes(hyaluronic acid,laminin, N-terminal procollagen, and collagen type IV) were determined by RIA and ELISA, and the degree of inflammation and fibrosis of liver tissues were examined by hematoxylin-eosin stain.Results Serum liver function parameters and fibrosis indexes improved more significantly in the oxymatrine-treated groups than those of the control group. The hepatic histologic examination showed more severe degree of hepatic inflammation and fibrosis in the control group than that in the oxymatrine-treated groups.Conclusion Oral oxymatrine can improve the serum biochemical and fibrotic indexes of rat liver fibrosis model induced by carbon tetrachloride, which suggests that oral oxymatrine has anti-fibrogenic effect.
