血管内皮细胞损伤机制研究 被引量:4

摘要 血管内皮细胞直接和血液接触,因此它很容易受到血液中活性物质的影响.1976年Ross与Glomset提出血管系统疾病内皮细胞损伤学说,开创了血管病原学研究的新领域.Ross在最初的理论中提出长期的高血压及血流冲击等剪切应力促使内皮损伤,引起血小板聚集及释放血管活性物质失衡,造成血栓形成及血管收缩;1993年又阐述了内皮细胞功能障碍学说,在动脉硬化的初始病灶上,单核细胞、T细胞结合粘连分子后,游走侵入内皮,并释放活性介质,引起中膜平滑肌细胞向内膜游走和增生.不正常的内皮细胞反应常伴随着器官功能的障碍,严重时会导致疾病的发生.虽然研究已证实,血管平滑肌细胞在高血压和动脉粥样硬化等血管内膜和中膜类的疾病中起重要作用,但由于内皮细胞可能是首先受血液中病理生理变化影响的细胞,多种因素的综合作用会导致内皮功能障碍,内皮细胞功能的改变可能是血管性疾病的重要环节,因此对血管内皮功能的研究有助于阐明许多血管性疾病的病理生理机制[1].
