复方苦参碱对人增生性瘢痕成纤维细胞的抑制作用 被引量:5

摘要 目的:观察复方苦参碱对人增生性瘢痕成纤维细胞的抑制作用,为烧伤后增生性瘢痕的治疗提供实验依据。方法:体外培养烧伤后的人增生性瘢痕成纤维细胞,加入不同含量的复方苦参碱,24h后观察细胞形态学,以乳酸脱氢酶(lactatedehydroge-nase,LDH)为指标观察细胞毒性,用四甲基偶氮唑蓝(MTT)法检测其增殖活性。结果:不同含量的复方苦参碱均能改变成纤维细胞形态,抑制细胞增殖,含量在300mg/L以下无明显的细胞毒作用,EC50值约为20mg/L。结论:复方苦参碱对人增生性瘢痕成纤维细胞具有抑制作用,并呈剂量依赖关系,具有防治增生性瘢痕的应用前景。 AIM:To explore the effects of compound prescription of sophocarpidine on postb urn hypertrophic scar-derived fibroblasts,so as to provide us an experimental s upport for the treatment of postburn hypertrophic scar . METHODS:Hypertrophic scar-derived fibroblasts were harvested and cultured in vitro.The proliferation activity of the fibroblasts was examined by Mymethyl-Th iazoldiphenyl Tetraziolum(MTT) method. The cellular morphology, proliferation ac tivity and the cytotoxicity of the drug were observed after 24 hours of culture. RESULTS:The fibroblast morphology could be altered by any concentration of com pound prescription of sophocarpidine solution. Furthermore, the cell count and t he cellular proliferation activity were all decreased obviously by the treatment of compound prescription of sophocarpidine.
