摘要
目的:探讨热诱导颊癌细胞凋亡过程中Bax基因蛋白及mRNA表达的动态变化,及其在细胞凋亡过程中的作用。方法:43℃水浴40min加热BcaCD885细胞,0,3,6,12,24,48h后收获细胞,采用流式细胞仪DNA蛋白质分析技术、RT-PCR技术检测诱导颊癌细胞凋亡过程中的凋亡率、Bax蛋白及mRNA表达水平。结果:43℃水浴40min后,凋亡率、Bax蛋白及mRNA表达明显升高,与对照组相比,有显著的差异性(P<0.05)。结论:高温作为一种外来刺激信号通过上调bax基因的表达,诱导BcaCD885细胞凋亡的发生。
AIM: To investigate the importance of the Bax gene in apoptosis induced by hyp erthermia in BcaCD885 cells. METHODS: Through hyperthermia at 43 ℃for 40min, apoptosis was induced in BcaC D885 cells. 0,3,6,12,24 and 48 hours after completing heating, the apoptotic rat e, the Bax protein and mRNA expressions of the cells were analyzed using flow cy tometry DNA analysis, flow cytometry protein analysis and reverse transcription -polymerase chain reaction analysis. RESULTS:After hyperthermia, the apoptotic rate, the Bax protein and mRNA expre ssions of the BcaCD885 cells there increased significantly.
