摘要
目的 :建立测定当归中阿魏酸含量的方法。方法 :采用毛细管电泳法 ,毛细管柱为 Beckman无涂层毛细管(57cm× 75μm,有效长度 50 cm) ,检测波长为 2 80 nm,进样压力 586k Pa,进样时间 6s,分离电压 2 0 k V,温度2 5℃ ,运行缓冲液为含 0 .2 mol· L- 1硼酸的 80 %乙醇溶液 (p H 9.4)。结果 :其线性范围为 10~ 53 0μg· ml- 1,回归方程为 Y=0 .0 2 0 2 X-0 .14 4 9,r=0 .9991,回收率为 99.6% ,RSD为 1.8% (n=6)。结论 :该方法简便 ,准确 ,重现性好 。
Objective: To establish a method for determination of f erulic acid in Angilica sinensis. Methods: The contents were determined by HPCE, with unt rea ted fused silica capillary (57 cm×75 μm ID, 50 cm effective length). Pressure injection was 586 kPa for 6 seconds, and the detection was performed at 280 nm. Running buffer was 80% ethanol solution containing 0.2 mol·L -1 boric aci d NaOH (pH9.4). The running voltage was 20 kV at 25℃. Results: The content of ferulic acid had a good lineari ty at the range of 10~530 μg·mL -1 , Y=0.020 2 X-0.144 9, r=0.999 1. The average recovery was 99.6% and RSD was 1.8% (n=6). Conclusion: The method is rapid, reliable and simple. It c an be used for the quantitative determination of ferulic acid in Angilica sine nsis.
