摘要
对分析植物激素用的高效液相色谱(HPLC)配置的色谱柱的半峰宽等特性进行了试验。用ODS类型的柱(长25cm),洗脱液在流量0.5ml/min时,当其中的甲醇浓度变更体积分数为φ=20%时,要15min样品保留时间才趋于稳定。对6种植物激素标准试剂,在不同的色谱柱中,洗脱液中的甲醇浓度与样品保留时间关系的试验表明,ODS类型的色谱柱有共同的溶出特性,二支柱串联和一支柱一样,具有相同的溶出特性。在甲醇浓度和样品保留时间取对数值所作的坐标图中,可得到很平缓的溶出特性曲线。注入的IAA量(溶质量)与峰面积有高度的正相关。当注入的甲醇量一定时,半峰宽也一定,而与IAA量(溶质量)无关。取半峰宽与洗脱液中甲醇浓度的对数值作坐标图,两者成直线关系。在样品半峰宽与保留时间的对数值坐标图中,4种植物激素(IAA,ABA,Z,ZR)都接近直线并且4条线紧密靠拢成一狭带。
Various properties such as halfband width of HPLC columns for phytohormone analysis were examined.As methanol concentration(φ=±20%) in eluent(0.5?ml/min) was changed using Octadecylsilane(ODS) type column in 25?cm length,the retention time became stable after 15 min.As to 6 standards of plant hormone,the relationship between methanol concentration in eluent and retention time was examined in each column.In ODS type column the general tendency of elution pattern was confirmed.As two columns were connected,the elution pattern was same as that of one column and no problem occurred.As in the elution pattern both methanol concentration and retention time was confirmed.As IAA amount (solute amount) and peak area were plotted,correlation was high and there existed a proportional relationship.Regardless of IAA amount injected,as injected amount of methanol was constant,the value of halfband width was constant.As halfband width and methanol concentration of eluent were plotted in log scale,they had an almost straight relationship.Moreover as halfband width and retention time were plotted in log scale,4 standards of phytohormone existed in a straight narrow belt.
