肝细胞生长因子对大鼠肝脏原位冷缺血再灌注损伤的保护作用 被引量:1

摘要 目的 观察大鼠肝脏原位冷缺血再灌注损伤的发生规律和肝细胞生长因子 (HGF)的保护作用。方法 建立大鼠肝脏原位冷缺血再灌注模型 ,分为保护组和对照组 ,分别于术前 30min给予HGF(1.5mg 10 0g体重 )和生理盐水 ,于术后不同时段处死大鼠并测定血清中的天门冬氨酸氨基转移酶 (AST)、丙氨酸氨基转移酶 (ALT)、透明质酸(HA)的浓度 ,检测肝脏匀浆中的丙二醛 (MDA)的含量。肝脏组织做光镜及透射电镜检查。并检测凋亡细胞数。结果 HGF保护组在各采样时间点上血清AST、ALT、HA和肝组织匀浆中MDA的含量均明显低于对照组 (P <0 .0 5或P <0 .0 1) ;HGF保护组中肝组织中凋亡细胞数低于对照组。结论 保护组的生化指标均明显低于对照组 ,复灌12 0min升高显著。HGF可减轻大鼠肝脏的冷缺血再灌注损伤。 Objective To study the protective effect of HGF on hypothermic is chemia-reperfusion injury. Methods Based on the model of liver hypothermic irrigation and repe rfusion,HGF(1.5mg/100g weight) and saline were separately given to the rat be f ore operation. The blood serum and hepatic tissue sample were collected to dete rmine the content of ALT,AST,HA and MDA at different time,the path ologic examinations of hepatic tissue were made by optical microscope. Results In the HGF group,the increse in serum alanine transaminase a nd hyaluronic acid level and the liver tissue MDA were significantly lower tha n that of control group(P<0.01 or P<0.05);the amount of apoptotic cell s were fewer than that of control group. Conclusion HGF can protect liver from cold ischemia-reperfusion in jury,HGF may be an effective pharmacological agent for prevention and treatment of hepatic cold ischemia-reperfusion injury.
