铜绿假单胞菌外毒素对淋巴细胞毒性作用 被引量:1

摘要 目的 探讨铜绿假单胞菌外毒素A(PEA)对人外周血单核细胞 (PBMC)的毒性剂范围及抗PEA血清对PEA的中和作用。 方法  采集健康成年人的静脉血 ,密度梯度离心分离PBMC。不同浓度的PEA与PBMC37℃、5 %CO2 共同孵育 4 8h ,用台盼蓝染色排除法和四甲基偶氨唑蓝 (MTT)法检测细胞存活率。 结果 台盼蓝染色排除法和MTT法结果一致 ,PEA在 10 0 0ng/ml时PBMC的存活率显著下降 (P <0 0 1) ,PEA剂量在 0~80ng/ml时 ,对细胞存活率无明显影响 (P >0 0 5 )。而抗PEA抗体完全中和PEA的细胞毒作用。 结论 PEA在 10 0 0ng/ml时 ,对PBMC表现出细胞毒作用 ,而抗PEA血清可以中和PEA的毒性作用。
