摘要
目的 用非线性混合效应模型法考察年龄、性别、体重、合并用药等对环孢素A清除率的影响。方法 收集临床 2 2 1例肾移植患者 32 3人次于肾移植后服用CsA达稳态的血药浓度谷值数据 ,应用非线性混合效应模型估算其清除率。结果 按口服一室开放模型计算CsA群体药动学参数 :清除率CL(L·h-1) =12 .81- 0 .0 6 7·年龄 +0 .10 1·体重。结论 患者体重、年龄对环孢素A清除率有显著影响。
OBJECTIVE: To estimate the influence of age, sex, body weight and concomitant medications on the relative clearance (CL) of cyclosporine A (CsA) in patients after renal transplantation using Nonlinear Mixed Effect Model (NONMEM). METHODS: 323 steady-state trough concentrations of CsA were collected from 211 patients. RESULTS. One compartment open model was used to analyze the data. The influence of the fixed effects on relative clearance of CsA was investigated using NONMEM. The final regression of CL (L · h -1) was: CL = 12.81 - 0.067·age + 0.101 · weight, where age was the age of patients (a), weight was total body weight (kg). CONCLUSION: Age and body weight influenced clearance of CsA in renal transplantation patients significantly.
