摘要
目的 研究中国北方人对肺癌的易感性与CyclinD1基因第 870位核苷酸A/G(A870G)多态性的关系。 方法 应用PCR SSCP方法 ,检测 93例肺癌及 12 2例健康对照外周血白细胞CyclinD1(A870G)的遗传多态性。结果 肺癌组与对照组CyclinD1(A870G)A/G的等位基因频率分别为 6 0 .75 %、39.2 5 %及 5 1.6 4 %、4 8.36 % ,两组间分布无显著性差异 (P =0 .0 6 )。肺癌组与对照组的A/A、A/G及G/G基因型频率分别为 38.71%、4 4 .0 9%、17.2 0 %及 2 3.77%、5 5 .74 %、2 0 .4 9%。两组间A/G及G/G基因型频率无显著性差异 (P >0 .0 5 ) ,而肺癌组A/A基因型频率明显高于对照组 (P =0 .0 18)。肺癌组与对照组A/A基因型频率的分布差异与肺癌的病理类型有关。肺鳞癌组的A/A基因型频率与对照组无显著性差异 (P =0 .4 0 3) ,而肺腺癌组的A/A基因型频率明显高于对照组 (P =0 .0 17)。与A/G及G/G基因型携带者之和相比 ,A/A基因型携带者患肺腺癌的校正相对风险度 (OR)为 2 .4 3,95 %CI =1.74~ 4 .77。结论 CyclinD1(A870G)A/A基因型是中国北方人患肺腺癌的危险因素 ,而肺鳞癌的易感性与CyclinD1(A870G)多态性无关。
Objective To investigate the association of Cyclin D1 (A870G) single nucleotide polymorphism (SNP) with susceptibility to lung cancer in northern Chinese population. Methods By polymerase chain reaction-single strand conformation polymorphism analysis (PCR-SSCP), the Cyclin D1 (A870G) genotyping was performed among 93 patients with lung carcinoma (50 adenocarcinoma and 43 squamous cell carcinoma) and 122 healthy controls. Results There were no significant differences in Cyclin D1 (A870G) allele frequencies between lung cancer patients and healthy controls(P=0.06). The A/A, A/G and G/G genotype frequencies were 38.71%,44.09%,17.20% respectively in lung cancer patients and 23.77%、55.74%、20.49% in healthy controls. There were no significant differences was A/G and G/G genotype frequencies between cancer patients and healthy controls (P>0.05). The A/A genotype frequency was significantly higher in lung cancer patients than in healthy controls(P=0.018)and this difference is related to the histology patterns. There was no significant difference of A/A genotype frequency between squamous cell carcinoma patients and healthy controls. However, the A/A genotype frequency of adenocarcinoma patients was significantly higher than that of healthy controls. The adjusted Odd Ratio of the A/A genotype was 2.43,95%CI=1.74~4.77 for adenocarcinoma patients. Conclusion In northern Chinese population, the Cyclin D1 (A870G) A/A genotype is a risk factor for lung adenocarcinoma. There is no significant association between squamous cell carcinoma and Cyclin D1 (A870G) polymorphism.
