摘要
用鼠细胞融合杂交瘤技术,获得3株分泌抗人T淋巴细胞表面抗原的单克隆抗体杂交瘤细胞株,分别为SMU_1、SMU_2、SMU_3,为IgG_1类,不与补体结合。间接免疫荧光法表明,SMU_1、SMU_3分别与50.3±6.42%和51.5±6.37%的周血单个核细胞反应,SMU_2与68.89±9.92%的周血半个核细胞及74.68±8.08%的成熟粒细胞反应,3株单抗的分子量为54~62kD(未还原)。调变试验及竞争抑制试验表明SMU_1、SMU_3与WuT_3识别相同的抗原分子,但为不同的表位。SMU_2为一非系限性单抗。本组单抗对研究T淋巴细胞分化及临床白血病免疫分型是有意义的。
In a fusion of BALB/c murine spleen cells immunizated with human fetal thymocytes and SP2/0 myloma cells,three monoclonaI antibodies (McAb)were produced,they were named SMU_1,SMU_2,SMU_3.They seem
to be new McAbs different from others.This paper describes their specificity which were analysied by indirect immunofluorescence technique.Results showed that SMU_1,SMU_3 reacted with peripheral blood mononuclear cells of 50.30±6.42% and 51.50±6.37% respectively.SMU_2 reacted with peripheral,blood mononuclear cells of 68.89±9.92% and mature granulocytes of 74.68±8.08%. None of these McAbs were reactive with red blood cells and platelets,their subtype are IgG_1 and not able to combine with completment,their molecule weight are from 54 to 62 kD.The modulation test and the competitive bin-ding assay suggested SMU_1,SMU_3 and Wu-T_3 reconized the same molecule,but different episodes.SMU_2 seems to be a non-linenage McAb.The group of
McAbs are useful for the study of T-lymphocyte differentiation and clinical leukemia classification.
