摘要
用台盼蓝染色及TNS荧光法测定光敏剂DE-03对癌细胞的光动力杀伤作用。结果表明,S_(180),Hepa,U_(14)癌细胞经DE-03 8μg/ml处理和黑光照射20min,杀伤率分别为65.0±2.5%,69.7±2.5%和81.3±2.2%,对S_(180)细胞50%的杀伤浓度为6.93μg/ml。向移植瘤小鼠静脉注射DE-03或YHpD20mg/kg后48h,氨离子泵浦染料激光照射20min,对小鼠S_(180)、U_(14)移植瘤的瘤重抑制率,DE-03为52.2%、47.8%,YHpD为31.3%、40.6%。以耳指数为观察指标,DE-03的光毒反应低于YHpD。
Photosensitizing agent DE-03 is a hematoporphyrin derivativ■separated from pig blood by our institute,It contains DHE 48.54%.The photody namic killing effect of DE-03 on cancer cells in vitro was studied by means of typan blue stained rate or TNS flourescent rate.The cancer cells pretreated with DE-03 or YHpD were exposed to black light for 20 min.The cellular killing rate of DE-03 at 8μg/ml to S_(180),Hepa and U_(14)was 65.0±2.5%,69.7±2.5% and 81.3±2.2% respectively.The 500% S_(180)cells killing concentration was 6.93μg/ml for DE-03 and 7.99μg/ml for YHpD,The photodynamic killing effect of DE-03 on transplanted tumors in mice was determined by means of inhibiting rate of tumor weight
and compared to YHpD.After intravenous injecton of DE-03 or YHpD at 20 mg/kg,the mouse transplanted S_(180)U_(14)were away from light for 48h.Then,the tumors were separably exposed to laser for 20min(wave length 630nm,power 190mV,diameter of light spot 1cm).After 6 days,the mouse were sacrificed and tumors were weight.The results showed that the inhibiting rate of tumor weight of S_(180)and U_(14)treated with DE-03 was
52.2% and 47.8% respectively,and treated with YHpD was 31.3% and 40.6% respectively.The pohtotoxic reaction of DE-03 was lighter than that of YHpD.
