细胞周期调控因子P16、Rb和cyclin D1在膀胱癌中表达的意义(英文) 被引量:4

摘要 目的 探讨细胞周期调控因子P16 -cyclin -Rb的通路与膀胱癌关系。 方法 应用免疫组织化学方法检测P16、Rb和cyclinD1在膀胱癌和正常膀胱组织中表达。 结果 P16和Rb在膀胱癌中表达显著低于正常组织 ,而且与肿瘤分级、分期密切相关 ,但与肿瘤复发无显著相关性 ;cyclinD1在膀胱癌中表达显著高于正常组织 ,但与肿瘤分级、分期和复发之间无显著相关性。膀胱癌中三种蛋白总异常率为 97.2 % ,其中同时两种或三种蛋白异常率达 6 1.1% ,肿瘤复发率也增高。膀胱癌中P16与Rb和cyclinD1分别呈负相关表达。结论 P16 -cyclin -Rb的通路异常与膀胱癌的发病机制密切相关 ,同时检测三者有助于更好了解膀胱癌生物学行动。 Objective:To investigate the expression of cell cycle regulators p16,pRb and cyclin D1 in bladder cancer and the relationships among these three proteins.Methods:Immunohistochemistry methods were adopted to examine expressions of p16,pRb and cyclin D1 in 72 cases of bladder cancer and 12 cases of normal bladder tissues.Results:Lower expressions of p16 and pRb were found in bladder cancer than those in normal bladder tissues(P<0.05), while both proteins expressions were closely associated, with tumor grade and stage(P<0.05). Expression of cyclin D1 in bladder cancer was significantly higher than that in normal bladder tissues(P<0.05),however, its expression was not significantly associated with tumor grade,stage and recurrence(P>0.05). The total abnormal percentage of p16,pRb and cyclin D1 in bladder cancer was 97.2%, of which that co-abnormal percentage of two or three was 61.1% and tumor recurrent rate increased in those cases.There were negative relatonships expression between p16 and pRb, p16 and cyclin D1.Conclusions:The abnormality of P16-cyclin-Rb pathway is highly related to the pathogenesis of bladder cancer. Co-examining these three proteins are helpful for evaluation of biological behavior and prognosis of bladder cancer.
