红麻花叶病病原病毒鉴定

IDENTIFICATION OF CUCUMBER MOSAIC VIRUS FROM AN AMBARY PLANT

摘要 从红麻花叶病株上分离到一病毒分离物(Amv-1),并可由汁液摩擦接种和由蚜虫传毒。人工汁液摩擦接种可侵染7科中的19种植物,系统侵染黄瓜、普通烟、心叶烟和番茄,局部侵染苋色藜、昆诺阿藜。该病毒钝化温度为55~60℃,稀释限点为10^(-3),体外存活期为3天(22℃)。用差速离心法可提纯病毒,部分提纯的病毒具有典型的核蛋白紫外吸收特性,最大吸收峰为258nm,最小吸收峰为239nm,A260/A280=1.68。SDS—不连续梯度聚丙烯酰胺凝胶电泳解离病毒,其病毒外壳蛋白亚基为一条多肽链,分子量约为26500道尔顿。琼脂免疫双扩散试验表明,该病毒与黄瓜花叶病毒抗血清具有阳性反应。经甲醛固定部分提纯病毒,1.5%磷钨酸负染,电镜观察病毒粒子球形,直径约为26nm。根据上述特征,该病毒分离物属于黄瓜花叶病毒的一个株系。 A virus isolate, Amv-1, was obtained from a diseased ambary plant with symptoms of mosaic, mottle and distortion. The isolate could be transmitted by sap inoculation to 19 species of 7 families and by aphides (Myzus persicea) in a non-persistent manner. The virus could systematically infected Cucumis sativus, Nicotiana tabacum, N. glutinosa, Lycopersicum esculentum and locally infected Chenopodium amaranticolar and C. quinoa. The thermal inactivated point ranged from 55 to 60℃ with life span in vitro being of 3 days (?)at 22℃) and the dilution end point of 10^(-3). The virus could be purified by differential centrifugation, and the partially purified virus had a typical ultraviolet absorption spectrum of nucleo-protein. The maximum and minimum UV absorption peaks were at 259 and 239nm, respectively, A260/A280=1.66. Only one chain of polypeptide with MW 26500 dalton appeared in discontinuous SDS-PAGE separation of the coat protein of Amv-1. In an agar gel double difusion test, the virus gave a positive reaction to antiserum of CMV. Fixed with 12% formaldehyde and negatively stained with 1.5% phosphotungstate, observed by electron microscopy, the partially purified Amv-1 particles were isometric, 26nm in diameter. Based on the results above, Amv-1 belongs to a strain of CMV.
