摘要
5-羟色胺(5-HT)是体内一种重要的生物活性物质,在神经系统中与递质功能密切相关,周围血中的5-HT除其有明显的血管活性外其它功能尚不清楚,但它的测定已广泛用于一些疾病的研究中。血液中的色氨酸(Trp)作为前体可影响5-HT代谢,同样与许多疾病有关;它以游离及结合两种形式存在,两者有不同的生理意义。其分离方法有透析、层析、超滤等。我们现报告一种用饱和硫酸铵沉淀蛋白以分离游离与结合色氨酸,用高效液相色谱(紫外及电化学检测器)测定5-HT及Trp的方法。
An HPLC method with electrochemical and ultraviolet detections to determine 5-hydroxytrytamine and tryptophan in blood has been developed. For separation of free and bound tryptophan , the serum sample was deproteinzed by the treatment with (NH4)2-SO4 saturated solution. The results of this method were similar to those of ultrafiltration, but the required sample volume was reduced from 30 ml to about 1. 5 ml. 5-HT content in platelet-rich plasma(PRP)sample was 10 times higher than that in platelet-poor plasma (PPP) sample. And 5-HT content in serum was positively correlated with that in PRP.
