摘要
目的 :观察缺血再灌注后早期皮层、尾壳核和海马细胞凋亡的变化。方法 :采用线栓法制成大鼠左侧大脑中动脉缺血再灌注模型(n =2 1) ,应用末端转移酶介导的dUTP -DIG缺口末端标记法 (TUNEL) ,标记凋亡细胞断裂的DNA。结果 :脑缺血再灌注后 12h ,左侧大脑中动脉缺血区皮层和尾壳核出现大量凋亡细胞 ,于再灌注后 2 4h~ 48h达高峰。结论 :神经细胞凋亡可发生于缺血再灌注后的早期。
Objective:To investigate the neurocyte apoptosis in the area of cortex, caudoputamen and hippocampus in the early stage after ischemic-reperfusion. Methods:The focal cerebral ischemia-reperfusion models of rats (n=21) were made with thread embolism of left middle cerebral artery(LMCA). DNA fragmentations of apoptotic cells were detected with terminal deoxynucletidyl transferase (TDT)-mediated dUTP-DIG nick end-labeling(TUNEL) method. Results:It was found that the apoptotic cells exhibiting DNA fragmentation were increased in the core ischemic area of cortex and caudoputamen of LMCA territory at 12h ischemia-reperfusion and peaked at 24h~48h of the reperfusion. Conclusion:The results indicated that the focal ischemia-reperfusion can induce apoptosis in early stage.
