摘要
目的 :评价凝血酶对血管内皮细胞通透性的影响及其机制。方法 :在体外培养血管内皮单细胞层 ,并测定凝血酶引起的血管内皮单细胞层电阻变化。结果 :凝血酶能够迅速降低血管内皮单细胞层的电阻 ;而肌球蛋白轻链激酶抑制剂 ML- 7能够抑制凝血酶引起的血管内皮单细胞层电阻变化。结论 :凝血酶可以升高血管内皮细胞肌球蛋白轻链的磷酸化水平 ,从而导致血管内皮细胞通透性增高。
Objective:To evaluate the effect of thrombin in endothelial cell permeability. Methods: A confluent HUVEC monolayer was cultured in vitro ,and the electrical resistance of a confluent HUVEC monolayer induced by thrombin was measured.Results:Thrombin reduced the electrical resistance of a confluent HUVEC monolayer rapidly When HUVEC was pretreated with the myosin light chain kinase inhibitor(ML 7),the changes of the electrical resistance of a confluent HUVEC monolayer that thrombin induced was inhibited. Conclusion:Thrombin stimulates phosphorylation of myosin light chain in endothelial cell and increases vascular endothelial cell permeability. [
