摘要
研究氡及其子体对大鼠外周血单个核细胞(Peripheral blood mononuclear cell,PBMC)、肺细胞、支气管肺泡灌洗液(Bronchi alveolar lavage fluid, BALF)细胞以及脾和胸腺细胞DNA的损伤作用。雄性SD大鼠吸入氡及其子体的累积剂量分别为66、111、174工作水平月(Working level month, WLM)后,收集BALF细胞,制备肺、脾脏和胸腺组织的单细胞悬液,采用单细胞凝胶电泳(Single cell gel electrophoresis,SCGE)技术,检测细胞DNA链的断裂改变。结果表明, 大鼠吸入氡及其子体后,各照射组PBMC、BALF和肺细胞DNA链断裂的迁移长度显著增加(p<0.01)。其中, PBMC与肺细胞DNA损伤有相关性(r =0.887, p<0.01),直线拟合(R2=0.787)的回归方程为Y=0.978X-0.12。在本实验的照射剂量下,氡及其子体能引起PBMC、BALF和肺细胞的DNA链断裂的损伤。
A study was done on DNA damage in tissue cells of ranon-inhaled rats. Sprague-Dawley rats were exposed to radon and its progeny to doses of 66, 111 and 174 WLM(working level month), respectively. Single cell suspensions of the rat lung, spleen and thymus were prepared. Single cell gel eletrophoresis(SCGE) was performed to detect the length of DNA migration in the rat peripheral blood mononuclear cells(PBMC), bronchi alveolar lavage fluids (BALF)cells, lung cells, spleen cells and thymus cells. DNA migration in the rat PBMC, BALF cells and lung cells increased with cumulative doses. The dose-effect relationships of the DNA damages and a correlation between DNA migration in PBMC and lung cells could be established. Radon and its progeny could cause DNA strand break in rat PBMC, BALF cells and lung cells.
