反义核酸对胃癌细胞端粒酶活性与细胞生长的抑制

摘要 目的 阐述反义核酸对端粒酶活性抑制作用的浓度依赖性和序列特异性,表明端粒酶活性表达对胃癌细胞分化的不依赖性。方法 运用改良的端粒酶活性定量检测法,测定三种胃癌细胞(MKN-28、SGC-7901、MKN-45)的端粒酶活性;用台盼蓝染色法,观察胃癌细胞的活力。结果 三种不同分化程度的胃癌细胞均有端粒酶活性的表达,但其活性的高低与其分化程度没有显著关系。在指定的浓度下,端粒酶反义核酸作用于胃癌细胞后,MKN-45和SGC-7901胃癌细胞出现明显的端粒酶活性和细胞生长的抑制,但在同样浓度条件下,MKN-28胃癌细胞只出现端粒酶活性的抑制。错义序列对照组则无明显变化。结论 端粒酶活性与胃癌细胞的分化没有显著相关性。端粒酶反义寡聚脱氧核苷酸能抑制胃癌细胞的生长和端粒酶活性,其抑制作用有浓度依赖性和序列特异性,其作用机制是通过抑制端粒酶活性,端粒酶活性的抑制并不总是同细胞生长的抑制平行的。 Objective In order to investigate the effect of specialized human telomerase anti-sense oligodeoxyribonucleotides on the growth and telomerase activity of gastric cancer cell lines under given circumstances. Methods Modified telomeric repeat amplification protocol was used to detect telomerase activity, using Typan Blue to explore the growth and activity of gastric cancer cell lines. Results Telomerase activity was detected in well, moderate and poor differentiated gastric cancer cell lines, but no association was found between telomerase activity and differentiation. Antisense human telomerase oligomers inhibited telomerase activity of all these gastric cancer cell lines. After treatment with antisense oligomers of different concentrations for 96h, growth inhibitory effect was also found in the MKN -45 and SGC -7901 gastric cancer cell lines. Conclusion Our results confirmed the former investigation with cytology that no statistic correlation was found between clincopathological parameters and telomerase activity. Antisense telomerase oligomers inhibited the growth and telomerase activity of gastric cancer cell lines,which depended on concentration and sequence specialty. The inhibition of telomerase activity is not always correlated with the inhibition of cell growth.
