抗纤灵冲剂对慢性肾衰模型肾组织TNF-mRNA、PDGF-mRNA的影响 被引量:33

The Effects of Kangxianling Decoction on PDGF-mRNA ,TNFα-mRNA Expression of CRF Rat Renal Tissue

摘要 目的 :观察抗纤灵冲剂对 5 6肾切除慢性肾衰 (CRF)大鼠肾组织肿瘤坏死因子α(TNF α)、血小板源性生长因子(PDGF)的影响。方法 :用RT PCR法检测CRF大鼠肾组织TNFα mRNA及PDGF mRNA表达及应用ELISA法检测实验后各组大鼠血液TNF α含量。结果 :正常组大鼠肾组织仅微弱表达PDGF mRNA、TNFα mRNA ,模型大鼠残肾组织有高表达 ,抗纤灵冲剂有较强的抑制作用 ,丹参、制大黄作用次之 ,蒙诺亦有一定的抑制作用。结论 :抗纤灵冲剂通过抑制肾组织TNFα mRNA、PDGF mRNA表达、降低血清TNF α含量 ,从而减轻了在肾小球和间质ECM的沉积 ,改善了肾脏纤维化 ,延缓了肾功能的恶化。 Objective:To observe the effect of kangxianling decoction on PDGF-mRNA,TNF α-mRNA expression of CRF Rat renal tissue. Methods:The expression of PDGF-mRNA, TNF α-mRNA level of CRF Rats renal tissue were determined by RT-PCR and the blood TNF-α content of CRF Rats was measured by ELISA method in all groups two months later. Results:There are high expression of PDGF-mRNA, TNF α-mRNA in renal tissue of the model group and there are slightly expression in false operation group. The kangxianling decoction could suppress significantly the expression of PDGF-mRNA, TNF α-mRNA level of CRF Rats renal tissue. The blood TNF-α content of model group was higher than those of the normal group (P<0.01). The kangxianling decoction could decrease significantly the blood TNF-α content of model group and its effect was better than those of Dan-shen root, Rhubarb and monopril groups (P<0.01).Conclusion:The kangxianling decoction could relieve the ECM deposition in renal glomerulus and interstitium, improve kidney fibrosis and retard the deterioration of renal function by suppressing the expression of PDGF-mRNA, TNF α-mRNA level of CRF Rat renal tissue, decreasing the blood TNF-α content and the effect of the PDGF, TNF α on cell proliferation of renal tissue.
