岩藻糖糖链在4-氨基苯酚维甲酰胺抑制肝癌细胞迁移中的作用

摘要 目的 观察N(4 氨基苯酚 )维甲酰胺 (4 HPR)对肝癌细胞迁移能力和细胞表面岩藻糖糖基化糖链表达的影响。方法 运用自行合成的 4 HPR ,一种经过羧基修饰的维甲酸衍生物 ,作用于具有高迁移能力的 772 1 k3肝癌细胞单克隆亚株。结果 经过 4 HPR 6h的短时期处理以后 ,① 772 1 k3细胞的穿膜迁移能力即受到显著抑制 ,下降至对照组的 2 7.1% ,抑制率与维甲酸作用相似 ;② 772 1 k3细胞与荆豆凝集素及小扁豆凝集素的结合率显著下降 ,分别下降至 91%和 87.5 % ,并与迁移力下降有一定的平行性。应用凝集素亲和层析柱分离提取了细胞膜中含有岩藻糖链的糖蛋白 ,用这些糖蛋白及其糖肽直接作用于 772 1 k3细胞 ,显示这些糖蛋白均有直接抑制肝癌细胞迁移力的作用 ,水解糖蛋白所产生的糖肽与糖蛋白作用相似 ,小扁豆凝集素吸附的糖蛋白在去除蛋白质部分以后 ,抑制作用有所加强。结论 与细胞迁移运动有关的岩藻糖基糖链可能参与了 4 HPR对肝癌细胞迁移行为的抑制。 Purpose: The purpose of this study is to observe the effect of N(4-hydrophenyl) retinamide (4-HPR) on fucosylated oligosaccharide expression on cellular surface, and the migration ability of hepatoma cells. Methods An artificial derivative from retinoic acid, which was synthesized by our group, was used to treat hepatoma cells 7721-k3 which was a sub-clone with a high mobility form SMMC-7721 hapatoma cell line. Results: It was observed that the migration ability of 7721-k3 cells was significantly inhibited after 6-hour incubation with 3 μmol/L 4-HPR and that the migration inhibition rate was as similar as those of all-trans retinoic acid. Cell surface fucosylation was at the same time investigated on such cells. A significant decrease in binding lectins of both ulex europaeus agglutinin (UEA) and lens culinaris agglutinin (LCA) in 7721 -k3 cells was noted after 6-hour treatment with 4-HPR, which seemed in parallel with the decrease of cell migration. Therefore, fucosylated glycoproteins were isolated by LCA and AAL lectin chromatography form the cell membrane and were directly added into the transwell chambers for migration assay to confirm the hypothesis that cell surface fucosylation might be involved in the migration inhibition by 4-HPR. In results, the isolated glycoproteins from LCA and AAL column were found significant inhibition of hepatoma cell migration. The subsequent glycopeptides digested from the glycoproteins inhibited the cell migration similarly. Such inhibition by exotic fucosylated oligosaccharides was dose-dependent, which suggested that the exotic oligosaccharides competed with cell surface sugar chains. Conclusions: The migration inhibition by 4-HPR in hepatoma cells might be associated with the reduced fucosylation in oligosaccharide chains on hepatoma cells.
