内异消治疗子宫内膜异位症实验研究 被引量:28

摘要 目的检测血管生成因子(VEGF)在子宫内膜异位症(EM)异位子宫内膜中的表达及肿瘤坏死因子(TNF-α)在EM腹腔液的表达,观察中药内异消对EM大鼠上述两因子的影响及治疗作用。方法建立EM大鼠模型,并随机分为内异消大、小剂量治疗组及丹哪唑治疗组、模型对照组、假模型组和空白对照组,检测VEGF在异位子宫内膜中的表达、TNF-α腹腔液中的表达。结果(1)模型对照组VEGF在异位内膜中的表达及TNF-α在腹腔液的表达显著高于假模型组;(2)内异消大、小剂量治疗组及丹哪唑治疗组VEGF在异位内膜中的表达强度及TNF-α在腹腔液的含量降低,与模型对照组比较,差异有显著性;(3)VEGF、TNF-α在EM时的表达呈正相关。结论VEGF、TNF-α均为血管生成因子,其促血管生成作用是EM发生发展的重要环节;内异消可通过调节TNF-α、VEGF的表达以抑制异位子宫内膜的生长。 Objective:To examine the expression of vascular endothelial growth factor(VEGF)in ectopic endometrium and tumor necrotic factor alpha (TNF-α)in peritoneal fluid of rates with endometriosis and to observe the effect of NeiYiXiao on two cytokines and therapeutic effect on experimental endometriosis .Methods:Implanted tunica intima method was used to develop animal model of endometriosis,NeiYiXiao was observed and compared with danazol,TNF-αconcentrations in peritoneal fluid of rates with ELISA,Immunochemical methods was used to determine the expression of VEGF in ectopic endometrium.Results:(1)VEGF expression in ectopic endometrium and TNF-αconcentration in peritoneal fluid of rates in ectopic endometrium and TNF-αconcentration in peritoneal fluid of rates in pathogenic model group were significantly higher than those of false model.(2)Neiyixiao could decrease the expressional intensity of VEGF in ectopic endometrium and the concentration of TNF-αin peritoneal floud of rats,there were significant difference as the drug treatment groups compared with the model group.(3)The expression of VEGF and TNF-αin the endometriosis showed direct propotion.Conclussion:As angiogenic factors,VEGF and TNF-αmay play an important role in the pathogenesis of endometriosis.NeiYiXiao may control the development of ectopic endometrium through adjusting the expression of VEGF and TNF-α
