摘要
目的 准确定位与老龄相关的豚鼠 m t DNA大片段缺失位点。方法 参照人类和大鼠线粒体 DNA大片段缺失的共同规律 ,运用计算机软件寻找豚鼠 m t DNA可能的缺失位点 ,采用聚合酶链反应 (PCR)、套式 PCR技术与 DNA测序技术 ,扩增豚鼠 m t DNA基因组可能的缺失区 ,并进行序列分析。检测耳蜗螺旋器组织、蜗神经组织、大脑颞叶组织中 mt DNA缺失的位点和片段大小 ,及 mt DNA大片段缺失的发生率。结果 首次发现豚鼠 mt D-NA在 nt86 36~ nt132 0 3位点之间存在 4 5 6 8个碱基的大片段缺失 ;老年豚鼠各组织中 mt DNA缺失率与青年组比较均有显著差异 ,前者 mt DNA缺失率明显高于后者。结论 豚鼠 mt DNA CO 与 ND5基因间有 4 5 6 8个碱基片段缺失。 mt DNA4 56
Objective To locate the mtDNA deletions of aged guinea pigs. Methods By reference to the common law of mtDNA deletions in humans and rats, the possible locations of mtDNA deletions in guinea pigs were examined by PCR and DNA sequencing. The cochlear Cortis, auditory nerve and brain tissues were harvested and the total DNA was extracted. Results It was discovered for the first time that there are 4568 bps deletions of mtDNA between nt8636 nt13203 in guinea pigs. The mtDNA deletion incidence of the aged group was significantly higher than that of the young control group ( P <0.05). Conclusion There exist 4568 bps deletions between COⅢ and ND5 genes of mtDNA in aged guinea pigs; mtDNA 4568 deletions of guinea pig possibly contribute to aging.
