肺纤维化大鼠肺泡灌洗液中碱性磷酸酶的动态变化及其意义 被引量:2

摘要 目的 :探讨碱性磷酸酶在肺纤维化中的作用。方法 :采用博莱霉素致肺纤维化大鼠模型 ,应用酶学方法对支气管肺泡灌洗液中碱性磷酸酶 (ALP)进行动态观察。结果 :(1)博莱霉素 (BLM )组支气管肺泡灌洗液(BALF)中细胞总数、中性粒细胞第 1d即开始增加 ,第 3d达高峰 ,14d后逐渐恢复正常 ;淋巴细胞在第 7d达高峰。 (2 )BLM组BALF中ALP的绝对值增加 (P <0 .0 5~ 0 .0 1) ;总蛋白第 3d达到高峰 (P <0 .0 1) ,第 14~ 2 8d逐渐恢复正常 ;ALP和总蛋白比率第 7d开始增加 (P >0 .0 5 ) ,第 14~ 2 8d逐渐达到高峰 (P <0 .0 5 )。血清中ALP两组无统计学差异。 (3)BALF中ALP绝对值与炎症细胞总分数以及总蛋白无明显相关 ,BALF中ALP和总蛋白比率与炎症细胞总分数也无明显相关。结论 :博莱霉素致肺纤维化大鼠BALF中ALP明显增加 ,ALP和总蛋白比率随着肺纤维化进展而增加 。 Objective: Our aim was to evaluate the role of alkaline phosphatase in pulmonary fibrosis. Methods: We investigated alkaline phosphatase (ALP) levels of BALF in different stages of bleomycin-induced pulmonary fibrosis in rats with enzymatic method. Results: In bleomycin group, the total inflammatory cells and neutrophils in BALF increased from the 1st day and reached the peak on the 3rd day. Another continue high level appeared from the 7th day to the 14th day. The lymphocytes reached the peak on the 7th day. In bleomycin-treated rats, absolute ALP levels in BALF were remarkably increased(P<0.05~0.01). Total protein reached the peak on the 3rd day (P<0.01), and gradually decreased to normal from the 14th day to the 28th day. Ration of ALP to total protein was increased from the 7th day(P>0.05), and gradually reached the peak from the 14th day to the 28th day(P<0.05). There was no significant difference in ALP levels in serum between the 2 groups. No correlation was found between ALP or ratio of ALP to total protein and BALF cytologic data. Absolute ALP level has no signigicant correlation with the total protein in BALF. Conclusion: In bleomycin-treated rats, ALP level in BALF was remarkably increased. Ratio of ALP to total protein ratio was increased with the development of fibrosis. The elevated ratio might be a marker of fibrosing progression.
