新化合物哌芳安他对血管内皮细胞氧化损伤的保护作用 被引量:3

摘要 目的 研究新化合物哌芳安他对培养的牛主动脉内皮细胞氧化损伤时ICAM - 1、MCP - 1mRNA表达水平的影响 ,探讨其抗动脉粥样硬化的作用机制。方法 使内皮细胞与不同浓度哌芳安他预孵育 2 0h ,再给予氧化低密度脂蛋白损伤 ,提取各组细胞总RNA ,通过逆转录 -聚合酶链式反应技术 ,以 β2 -微球蛋白为内参照检测内皮细胞ICAM - 1、MCP - 1mRNA表达水平的变化。结果 内皮细胞与哌芳安他预孵育后 ,再受到氧化损伤时 ,其ICAM - 1、MCP - 1mRNA的表达水平明显低于与ox -LDL直接接触的细胞。结论 哌芳安他在一定程度上可保护内皮细胞使之免于或减轻氧化损伤。 Aim To investigate the effects of pivanampeta against ICAM-1 and MCP-1 mRNA expression in cultured endothelial cells derived from bovine aorta.Methods The ICAM-1 and MCP-1 mRNA expression in bovine aorta endothelial cells was measured by reverse transcription -polymerase chain reaction and normalized to the mRNA level of β 2 -microglobulin in the absence or in the presence of ox-LDL or land pivanampeta Results Both ICAM-1 and MCP-1 mRNA expression in the endothelial cells derived from bovine aorta were increased by ox-LDL stimulation,which could be prevented by pivanampeta at the concentrations of 10 and 100μmol·L -1 .Conclusions The over expression of ICAM-1 and MCP-1 in endothelial cells induced by ox-LDL could be blocked by pivanampeta.Pivanampeta can protect the endothelial cells from the oxidative injury.
