米非司酮抑制胃癌生长及侵袭转移的研究 被引量:8

Effects of mifepristone on the growth and metastasis of human gastric cancer in vivo and in vitro

摘要 目的 探讨米非司酮 (MIF)对人胃癌细胞株SGC 790 1生长和侵袭转移潜能的影响及其作用机制。方法 应用原位杂交法检测胃癌细胞孕激素受体 (PR)的表达 ;Transwell膜侵袭系统分析MIF对胃癌细胞迁移潜能的影响 ;建立裸鼠胃癌移植瘤模型 ,给予MIF(5 0mg·kg-1·d-1)皮下注射治疗 6周后 ,检测胃癌组织生长和转移情况 ,以及微血管密度 (MVD)、血管内皮生长因子(VEGF)、增殖细胞核抗原 (PCNA)和半胱天冬酶 3 (Caspase 3 )表达的变化。结果 SGC 790 1细胞高表达PRmRNA。经 2 0 μmol/LMIF作用 2 4h后 ,胃癌细胞的迁移细胞数 (5 6± 5 )明显低于对照组 (10 4± 12 ,P <0 .0 1)。MIF显著抑制裸鼠胃癌移植瘤的生长 (抑瘤率为 41.15 % )和肺转移灶的形成 ,下调VEGF、PCNA的表达及MVD ,同时上调表达Caspase 3。结论 MIF能有效抑制胃癌生长和侵袭转移 ,其作用机制可能与抑制胃癌细胞运动、新生血管形成。 Objective To explore the effects of mifepristone (MIF) on the growth and metastasis of the SGC-7901 human gastric cancer cells in vivo and in vitro.Methods The expression of progestin receptor (PR) in SGC-7901 cells was detected by using in situ hybridization assay.A membrane invasion culture system,Transwell Insert,was used to analyze the migrative potential of the cells.SGC-7901 cells were subcutaneously xenografted in nude mice.Mifepristone was administrated for 6 weeks at a dose of 50 mg/kg daily to explore its effects on the tumor growth and metastasis,and the expression of microvessel density (MVD),vascular endothelial growth factor (VEGF),proliferating cell nuclear antigen (PCNA) and Caspase-3 detected by immunohistochemical staining.Results PR mRNA was highly expressed in the SGC-7901 cells.After treatment for 24 h with 20 μmol/L MIF,the number of the migrated SGC-7901 cells (56±5) was significantly deceased as compared with control group (104±12,P<0.01).MIF could markedly inhibit the tumor growth with the inhibitory rate being 41.15 %.Lung metastasis was seen in 3 mice of control group but in only one mice of the MIF-treated group.In addition,MIF could down-regulate the expression of MVD,VEGF and PCNA,whereas up-regulate the expression of Caspase-3 in gastric cancer tissue.Conclusion MIF can effectively inhibit the growth and metastasis of gastric cancer cells in vitro and in vivo,which may be related with the decrease of tumor cell proliferation,migration and angiogenesis,and the simultaneous increase of tumor apoptosis.
