膀胱癌细胞p16抑癌基因失活的研究 被引量:2

摘要 目的 探讨膀胱癌中 p16基因失活情况。 方法 采用PCR、PCR SSCP和DNA甲基化分析技术等方法对 2 5例膀胱癌组织中 p16基因进行检测。 结果  2 5例膀胱癌标本中有 6例存在 p16基因纯合性缺失 ,缺失率为 2 4 % ;无 1例出现点突变 ;16例出现异常甲基化 ,检出率为 6 9.6 % (16 /2 3)。结论 外显子 2的纯合性缺失在安徽区域发生的膀胱癌中可能不是一个较频繁发生的事件 ;通过CpG岛异常甲基化失活可能是膀胱癌 p16基因失活的主要机制。 Objective To investigate the inactivation of p16 gene in bladder cancer. Methods The p16 gene was analysed by duplex PCR,PCR SSCP and DNA methylation analysis in 25 bladder cancer tissue samples. Results The gene deletions were found in six samples, the deletion ratio of p16 gene was 24%. No case showed point mutation; 16 cases showed p16 gene 5′ CpG island methylation. Conclusion The deletion of p16 exon 2 is not a frequent event; a statistically significant association is also found between p16 gene 5′ CpG island methylation and the inactivation of p16 gene in bladder cancer.
