Bcl-2硫代反义核酸(G3139)OPC纯化方法

摘要 目的 建立 Bcl- 2硫代反义核酸 (G31 39) OPC纯化方法。方法 将 G31 39从合成柱上解离并去保护 ,用 OPC柱分离纯化化学合成的 G31 39,紫外分光光度计测 OD值 ,2 0 % PAGE鉴定其纯度。结果 经 OPC柱纯化后 G31 39制备量可达 mg水平 ,回收率为 6 6 % ,电泳结果显示为一条亮带。结论  Bcl- 2硫代反义核酸 OPC纯化法是一种高效、快速。 Objective\ To develop a method of OPC purification of antisense phosphorothioate oligodeoxynucleotide to Bcl 2.Methods\ After completion of the trityl on synthesis,G3139 was cleaved from the DNA synthesis column and deprotected,G3139 was purified by OPC column and its concentration was determinated by OD(A260),The purity of G3139 was analyzed by 20% PAGE method.Results\ The yields of purified G3139 could reached mg level and the productive rate being 66%,A single band was obseved from the result of 20% PAGE.Conclusion\ OPC colum provides a high effciency,quick and convenient method for the purification of antisense phosphorothioate Oligodeoxynucleotide to Bcl 2.
