摘要
本文介绍二氢玉米素核苷(DHZR)组的间接酶联免疫吸附测定法的研制结果。用过碘酸盐氧化法合成免疫原,获得的抗 DHZR 抗血清效价为1∶3090,该抗血清基本上只与 DHZR 组发生交叉反应.检测极限为14 fmol DHZR/孔,检测线性范围为0.1~100 pmol DHZR/孔。板内误差 C.V=1.8%,板间误差C.V.=5.8%,重复性较好。健全性试验表明样品测定值可信。并对玉米籽粒等试样中的 DHZR 组细胞分裂素含量变化作了测定。
The indirect enzyme-linked immunosorbent assay(ELISA)specifically for the determinationof dihydrozeatinriboside(DHZR)-group cytokinins(DHZRs,including DHZ,DHZR and DHZR-5'P)has been developed.The immunogen DHZR-N_9-BSA has been synthesized with NalO_4,oxidation.Theanti-DHZR antiserum has been raised in rabbit and its titer,determined with HRP-linked Protein A(PPA),reaches 1:3090.This antiserum cross-reacts only with DHZR-group cytokinins.The detection.lim-it of this ELISA is 14 fetomol DHZR/well and its linear detection range is 0.1~100 picomol DHZR/well.The coefficient of variation within microtiter plates is 1.8%,and that between microtiter plates is 5.8%. Thevalidity test indicates that the data of DHZRs contents in plant samples are correct.
