摘要
以纯化的牛冠状病毒(BCv)免疫 BALB/c 小鼠,取其脾脏细胞与小鼠骨髓瘤细胞 SP2/0融合,用间接 ELISA 筛选,获得了4株分泌 BCV 特异性单克隆抗体的杂交瘤细胞:C_(12)、C_(31)、C_(42)和C_(54).它们系针对 BCV 不同表面抗原决定簇的.用 C_(12)和 C_(31)两株细胞制备的腹水混合包被聚苯乙烯微孔板,建立了以生物素标记的兔抗 BCV IgG 和酶标亲和素介导的 BAS—ELISA,并对 BCV 标准毒株和临床犊牛腹泻粪样作了检测.
Myeloma cell line SP2/0 was fused with spleen cells from mice immunized with purifiedbovine coronavirus(BCV MUC 270)and 4 hybridoma cell strains(C_(12),C_(31),C_(42)andC_(54))secretingmonoclonal antibodies(McAbs)specific for BCV were obtained by screening with indirect ELISA.The char-acteristics of the four McAbs were identified.Microtitration plates were coated with McAb C_(12) and C_(31) in combination and a BAS-ELISA for de-tecting BCV antigen was established by use of anti-BCV rabbit IgG biotin conjugate and avidin peroxidaseconjugate.BCV in cell culture and in thirty feacal samples from calves with diarrhea were detected byBAS-ELISA,as well as HA/HI.It was suggested that BAS-ELISA could be used for the detection of BCVin veterinary clinic.
