摘要 目的对大肠艾希菌(E.coli)进行随机扩增多态性DNA(RAPD)基因分型并应用于医院感染的判断。方法以优化的随机引物、反应体系和扩增条件对临床分离的161株E﹒coli进行RAPD基因分型并按指纹图上DNA条带数及片段大小绘制基因分型图谱,同时对E.coli的医院内感染及个体感染的连续性进行了观察与分析。结果161株临床分离E.coli共得RAPD120型;E.coli的医院内感染确实存在。结论RAPD可将E.coli分为120型并有效应用于E.coli医院内感染的判断。 Objective To type Escherichia coli(E.coli)with randomly amplified polymorphic DNA(RAPD)analysis and judge the nosocomial infection.Methods Optimal random primer and reaction system and amplification condition of RAPD were used for genotyping of the161strains E.coli isolated from the clinical samples and the genotype map was drawn according to the numbers and lengths of DNA frag-ments.At the same time,nosocomial infection and individual infection continuity of E.coli were observed and analyzed.Results The161strains E.coli could be distinguished into120kinds of RAPD type and there were nosocomial infection indeed in hospital wards.Conclusion RAPD could genotype the E.coli into120kinds,and be applied to judge the nosocomial infection caused by E.coli effectively.
