摘要
本文应用异型双功能基化合物SPDP与单克隆抗体反应,产生单克隆抗体-PDP,然后再与暴露巯基的RTA结合,制备肝癌单抗-RTA结合物。ELISA及毒素重组试验鉴定表明:结合物在10^(-9)mol/L浓度时与靶细胞仍有较好的结合能力,结合物中RTA保持良好活力。体外细胞毒结果表明:结合物对靶细胞具有良好的导向杀伤作用,在10^(-9)mol/L浓度时对人肝癌细胞株BEL7404的杀伤率为41.2%,对正常细胞SL_7的杀伤力很弱。
In the present study,a conjugate of murine monoclonal antibody against hepatic cancer cell (Hepl)and ricin A-chain(RTA) was obtained by covalently coupling RTA to monoclonal antibody ,which was modified by introduce SPDP to 3-(2-pyridithio) propiony radical and then reacted with thiolated RTA. The conjugate obtained retained well the activity of antibody and the biological activity of A chain. The cytotoxic assay showed that the conjugate had a selective killing to human hepatic cancer cell line BEL_(7404), which 41. 2% BEL_(7404) cell was killed at a concentration of 10^(-9)mol/L,the conjugate was no toxic to. SL_7.
