摘要
本文建立了人血清胰岛素固相“夹心”时间分辨免疫荧光测量法。采用多克隆抗体包被微量滴定条,制成固相抗体。另将纯化的多抗与生物素偶联,制得生物素化抗体。Eu^(3+)标记链亲和素为示踪物。本法曲线剂量范围为1.25~320μIU/ml,最小检出值为0.5μIU/ml,平均批内及批间C.V分别为8.04和9.16%,平均回收为97.44%。本法快速方便,适于大量样品检测。与RIA相关性良好。
The article descripes a time-resolved fluoroimmuno-assay (TRFIA) for human serum insulin, based on the use of poly clonal antibody and biotin-strep-tavidin system (BAS) . The polyclonal antibody is immobilized on the surface of microtiter strip wells. The purified antibody is biotinated. And Eu^(3+)-ions are bound to streptavidin as the tracer. The sensitivity of the assay is 0.5μIU/ml, with a working range of 1.25-320μIU/ml. The mean intra and inter C.V of the assay are 8.04% and 9.16% respectively. The mean recovery is 97.44%. The assay is easy and rapid to perform and correlate well with RIA.
