摘要
应用淋巴细胞杂交瘤技术,建立了3株抗人血清载脂蛋白AI杂交瘤细胞株并制备了单克隆抗体(McAb),对其免疫学特性进行了研究。3株McAb均为IgG2a,效价分别为2.0×10^(-7)、1.0×10^(-3)和2.5×10^(-3),与其它载脂蛋白及白蛋白没有交叉反应。通过单抗相加试验及双抗体竞争试验的结果,证实3株McAb是识别载脂蛋白A两个不同的抗原决定簇。本文还对McAb的亲和力进行了测定。
Three hybridoma lines (1A_6, 5C_2, 6C_1) had been established, which are able to secrete IgG2a class monoclonal antibodies against human serum apolipopro-tein A I . Their immunological characters had been studied. The titers of these monoclonal antibodies were 2.0×10^(-7), 1.0×10^(-8) and 2.5×10^(-8) respectively. No cross reaction with apolipoprotein AⅡ, B CⅠ, CⅡ, CⅢ, E and albumin was noticed. Through monoclonal antibodies additivity test, calculations of additivity index and ELISA double antibody competive assay, it was shown that these monoclonal antibodies are against two antigenic determinants of apolipoprotein A Ⅰ . The affinity of three monoclonal antibodies were determined by ELISA with the results of 0.9μg/ml, 0.6μg/ml, and 0.2μg/ml respectively.
