肌肉标志结蛋白单抗的研制与初步应用 被引量:1

摘要 以鸡胗为材料提纯结蛋白,经鉴定后免疫BALB/c小鼠,采用杂交瘤技术建立了能稳定分泌抗结蛋白单抗的杂交瘤细胞株AD1,其染色体中位数为77。AD1单抗属小鼠Igg1亚类;ELISA间接法测定AD1细胞培养上清抗体效价为10^(-4);酶联免疫印迹试验显示能特异地与结蛋白起反应;经ABC-AEC免疫酶组化法鉴定组织特异性,AD1单抗具有特异地识别肌肉组织和肌源性肿瘤的能力,能用于常规病理标本的石蜡切片。AD1单抗对肌源性肿瘤和非肌源性肿瘤的病理诊断与鉴别诊断具有重要应用价值。 BALB/c mice were immunized with desmin isolated from chicken gizzard. Hybridoma cell line (AD1) continuously secreting anti-desmin monoclonal antibody was established by the hybridoma technique. The median of AD1 s chromosome is 77. AD1 McAb is of IgG1 subclass and its titre in cuture is 10. It reacts only with desmin in immunoblotting and specifically recognizes muscle tissue and myogenic tumors immunohistologically in conventional paraffin embedded sections.This indicates that ADl McAb is valuable for the diagnosis and differential diagnosis of myogenic tumors and non-myogenic tumors in pathologic specimens.
