摘要
本文应用DNA-磷酸钙共沉淀法及电脉冲介导基因转移法将人IL-2受体基因转染Amphotropic病毒包装细胞系-PA317细胞,比较了两种转移方法对逆转录病毒载体的转染效率。实验结果表明,电脉冲介导逆转录病毒载体的转染效率高于磷酸钙沉淀法;并研究了逆转录病毒载体导入病毒包装细胞的部分电场参数对基因转染频率的影响及最适的电转移条件。
Human IL-2 receptor gene was trans fected into Amphotropic viral packaging cell-PA317 cell line by electroporation and calcium phosphate-DNA coprecipitition methods. The results demonstrate eletric field mediated gene transfer is a more efficient technique than the calcium-phosphate method for transfection frequency of retrovi-ral vector in these cells. In the present study several electrical parameters affecting transfection of retroviral vector into viral packaging cell and optinum conditions were evaluated.
