摘要
为寻求一种检测鸭出血症病毒 (duck hemorrhagic disease virus,DHDV)快速、敏感、实用的方法 ,建立了间接免疫荧光试验 ,并测定了一抗 (兔抗鸭出血症病毒阳性血清 )、二抗 (FITC标记的羊抗兔 Ig G)的最佳使用浓度及最佳感作时间分别为 1∶ 2 0、6 0 m in和 1∶ 10 0、6 0 min。以建立的间接免疫荧光试验检测了 DHDV于番鸭胚成纤维细胞中的复制动态 ,结果表明 ,接毒后 36~ 6 0 h为 DHDV大量复制期 ;6 0~ 12 0 h为 DHDV从核膜以出芽方式进入胞浆中成熟的阶段 ;12 0~ 14 4 h为大量 DHDV自胞浆中释放至胞外阶段。该方法快速、特异 ,可用于该病的实验室诊断。
Indirect immunofluorescence assay has been developed for experimental diagnosis of the duck hemorrhogic disease.The optimal concentration of rabbit anti DHDV positive serum was determined to be 1∶20 ,the optimal reaction time to be 60 min.The optimal concentration of FITC(fluorescein isothiocyanate) labelled sheep anti rabbit IgG was determined to be 1∶100 ,the optimal reaction time to be 60 min.The test showed this assay is practical,quick and specific.The replication dynamics of DHDV in muscovy duck embryo fibroblast was observed under fluorescence microscope by using indirect immunofluorescence assay.The result indicated that replication of DHDV is major within 60 h post inoculation(PI),release of DHDV from cell nucleus to cell plasma is during 60 to 120 h PI,release of enormous DHDV out of cell begins at 120 h PI.
