摘要
目的:探讨在食管鳞癌中Survivin蛋白的表达及其与Bcl-2的表达和肿瘤细胞凋亡的关系。方法:用免疫组织化学染色法检测了64例食管鳞癌组织Survivin和Bcl-2的表达,同时用原位末端标记法(TUNEL)检测肿瘤细胞的凋亡,并进行了相关性分析。结果:68.7%的食管鳞癌表达Survivin蛋白,而正常食管上皮组织未见表达。Survivin的表达率与肿瘤的分期、分化程度和淋巴结转移无明显相关。40.7%的食管鳞癌同时表达Survivin和Bcl-2,二者存在显著相关。表达Survivin和/或Bcl-2的食管鳞癌的细胞凋亡均显著减少。结论:食管鳞癌细胞表达Survivin蛋白,在食管鳞癌的发生和发展中可能起重要作用;Survivin选择性表达于食管鳞癌的肿瘤细胞中,为食管鳞癌的靶向治疗提供方向。
Purpose: To study the expression of Survivin and Bcl-2 and their correlation with tumor cell apoptosis in esophageal cancer. Methods: The expression of Survivin and Bcl-2 was detected by immunohistochemical assay in 64 cases of esophageal cancer. Tumor cell apoptosis was identified and quantified by the terminal deoxynucleotidyl transferase-mediated dTUP nick and labeling( TUNEL) method. Results: The Survivin was expressed in 38 of 64 (68. 7%) cases of esophageal cancer of histological grade 2 to 3. In contrast, normal esophageal epithelium did not express Survivin. Survivin-positive cases were associated with Bcl-2 expression and reduced apoptotic index . The expression of Survivin was not related with histological grade, lymph node metastasis and histological type. Conclusions: The expression of Survivin in esophageal cancer, alone or in cooperation with Bcl-2, may play an important role in tumorigenesis through inhibition of apoptosis. The selective distribution of Survivin in tumor cells may provide a new direction for targeting therapy.
