摘要
目的 建立反相高效液相色谱法测定人血清中多索茶碱浓度。方法 采用高效液相色谱法 ,苯巴比妥为内标 ,YWG-C1 8色谱柱 ,甲醇∶水 ( 3 0∶ 70 )为流动相 ,流速 1.0 ml·min- 1 ,柱温 3 5℃ ,检测波长 2 10 nm。 结果 多索茶碱在 0 .40~ 2 4.0 0 μg· m L- 1浓度范围内呈良好线性关系 ,r=0 .9997;血清最低检测限为 10 0 mg·m L- 1 ;平均回收率为 98.71%;日内和日间 RSD分别为 2 .2 9%和 3 .3 7%。 结论 本法灵敏简便、准确可靠 。
OBJECTIVE To establish a method of RP HPLC for the determination of doxofylline concentration in human plasma.METHODS An RP HPLC was carried out on YWG C 18 .Phenobarbital was used as the internal standard.The mobile phase was composed of methanol water(30∶70),the flow rate was 1 0ml·min -1 the column temperation was set at 35℃,the detecting wavelength was set at 210 nm.RESULTS The assay was shown to produce linear calibration curves over the range from0 40μg·ml -1 to 24 00μg·mL -1 for doxofylling,r=0 9997,and the lowest plasma limitation was 100mg·mL -1 .The average recovery was 98 71%and the coefficients of variation within a day and between days did not exceed2 29%,3 37%.CONCLUSION The method is found to be sensitive simple convenient and accurate for the determination of doxofylline concentration in human plasma and clinical pharmacokinetic study.
