摘要
Caenorhabditiselegans是发育生物学中的重要模式生物,也是全基因序列已知的唯一多细胞动物.许多基因参与C.elegans的细胞命运决定和细胞谱系发育.mex 3是C.elegans早期胚胎发育中细胞命运决定的重要基因.本文报告采用原位杂交技术检测mex 3mRNA在C.elegans野生型胚胎发育中的分布,发现mex 3mRNA在生殖腺和卵母细胞的细胞质都有广泛而丰富的分布,从受精开始mex 3mRNA的分布开始发生了变化,从均一分布的模式开始进入不对称的分布模式,在2细胞阶段mex 3mRNA仅在前部的大卵裂球分布很多、染色较深,后部的小卵裂球低水平分布,染色较浅,在4细胞的早期胚胎中只在前部的两个卵裂球有较丰富的分布,后部的2个细胞低水平分布.到8细胞和16细胞阶段,只有后部的一个卵裂球检测到mRNA的分布,此后的胚胎中未检测到mRNA的分布.
C.elegans is an important model animal in developmental biology, and it is the only one multicellular organism with the completion of the genome project. A lot of genes involve in C. elegans cell fate determination and linage development. In C.elegans early embryo development, mex3 is an important gene involved in C.elegans cell fate determination and early embryo development. Its distribution and expression pattern give an impact on the other gene distribution and cell fate. Here we reported mex3 mRNA distribution pattern changes with the time in C.elegans wild type embryo via in situ hybridization. In wild type embryo,mex3 mRNA has a abundant distribution in oocyte and one cell embryo. From 2cell embryo, the mRNA distibution became different. The anterior blastomere express high level than the posterior cell, and same situation happened in 4cell embryo, mex3 mRNA in 2 anterior blastomeres presents high level, low level shows in 2 posterior cells. In 8cell and 16cell embryo, only one posterior blastomere shows mex3 mRNA.
