BDNF对小鼠周围神经损伤后髓化与再生作用的实验研究 被引量:9

摘要 目的 :建立小鼠坐骨神经压榨损伤模型 ,研究内源性的脑源性神经营养因子 (BDNF)对周围神经损伤后髓化与再生的作用。方法 :将动物分为三组 ,腹腔分别注射抗BDNF血清、羊血清和生理盐水 ,采用电子显微镜观察周围神经损伤后髓化与再生的变化 ,用体视学方法计数单位面积髓化纤维的数密度。结果 :抗BDNF血清组动物 ,髓鞘松散、分离、变性 ,损伤远端髓化轴突的数密度比羊血清组和生理盐水组分别减少 83%和78%。结论 :周围神经损伤后神经纤维的髓化和再生可能与内源性BDNF的作用有关。 Objective:The present study was to investigate effects of endogenous BDNF (brain-derived neurotrophin factor) on myelination and regeneration of injured peripheral nerve.Methods:Mice was intraperitoneally injected anti-BDNF,goat normal serum and 0.85% NS respectively.Electron microscope and stereology were employed.Results:The myelinated axons of anti-BDNF treated mice became shrunden,collapsed and degenerated;laminal layers of the myilin sheath became loosened and separated.In the distal side to lesion ,the quantities of myelinated axons were substantially reduced to 83% ,78% compared to goat serum treated and NS teated groups.Conclusions:Endogenous BDNF is required for remyelination and regeneration of injured peripheral nerve.
